Purification and Immunobiochemical Characterization of Sheep Hydatid Cyst Fluid Antigen

چکیده
In the present study, sheep crude hydatid cyst fluid antigen (SCHCFA) was prepared by ammonium sulfate precipitation, centrifugation and dialysis. SDS-PAGE of SCHCFA showed seven polypeptides of 72.8 kDa, 66.5 kDa, 60.2 kDa, 41 kDa, 25.1 kDa, 19 kDa and 8 kDa when stained with Coomassie briliant Blue R-250. The initial part of the ascending loop of the first peak (PI) when resolved by gel filtration chromatography on Sephacryl S 200 depicted the well defined polypeptide of molecular weight of 66.5 kDa and 60.2 kDa showed on 12.5% SDS-PAGE. Double immunodiffusion test, indirect ELISA and western blot analysis demonstrated that the 66.5 kDa and 60.2 kDa polypeptides were immunoreactive when treated against hyperimmune sera and known positive sera. It was concluded that these polypeptides (purified sheep hydatid cyst fluid antigen) might prove to be a promising tool for the diagnosis of hydatid disease.

A NOVEL METHOD OF PURIFICATION OF SPECIFIC HYDATID CYST ANTIGEN
Hydatid cyst fluid has been used as a source of antigen for the serodiagnosis of Echinococcus granulosus infection. Due to cross-reactions with antigens shared by other helminthes, the specific antigen from hydatid cyst fluid was purified by many workers. We used a relatively simple technique for purification of specific antigen from sheep hydatid cyst fluid. Isopycnic ultracentrifugation w...

○ INTRODUCTION Hydatidosis is a silent, cyclozoonotic and oldest infection of man and domestic animals and is caused by larvae of the cestode, Echinococcus (Bhatia and Pathak, 1990). The infected human beings and animals loose their normal functional capacities leading to huge economic losses. The diagnosis of hydatid disease is based on clinical, radiological, microscopical and immunological m...
متن کاملIsolation and purification of Echinococcus granulosus antigen B from hydatid cyst fluid using three different methods
Hydatid cyst, the larval stage of cestodes Echinococcus spp., is recognized as a zoonotic infection in the world. The World Health Organization (WHO) has recently classified echinococcosis in a group of neglected tropical diseases. The prevalence of Echinococcus granulosus infection is high in Iran due to the presence of various intermediate hosts in this country. Considering the rising trend of...
متن کاملIdentification and purification of a specific and immunogenic antigen of the laminated layer of the hydatid cyst and production of an antigen-specific monoclonal antibody.
Cystic echinococcosis (CE) is an infection caused by the larval stage of Echinococcus granulosus. This is widely distributed through Iran, where a variety of animals act as intermediate host. The immunogenic antigens (Ag) of different compartments of the hydatid cyst have been already determined. One of these compartments is the laminated layer (LL). We have extracted a protein with the MW of 2...
متن کاملEvaluation of Enzyme Linked Immunosorbent Assay, Utilizing Native Antigen B for Serodiagnosis of Human Hydatidosis
Background: Hydatidosis is one of the cosmopolitan parasitic zoonoses caused by the larval stage of Echinococcus granulosus. Diagnosis of hydatidosis is still an unresolved problem. Serological tests using crude antigens for diagnosis of E. granulosus are sensi-tive, however their specificity are not satisfactory. Therefore, WHO recommended spe-cific serological methods using specific antigens,...
